In microscopy, Leishman Stain is used to stain blood smears. It has a great stain quality. It is most commonly used to distinguish and identify leucocytes, malaria parasites, and trypanosomes. It is based on a methanolic combination of methylene blue that has been ‘polychromed’ (i.e. demethylated into different azures) and eosin. The methanolic stock solution is stable and also helps to directly fix the smear, removing the need for a prefixing step.
The Leishman stain typically displays the beautiful violet color of the nucleus and neutrophil granules, making differential count easier and improving staining quality over basic methylene blue and Eosin stains, which do not provide adequate contrast between the cytoplasm and the nucleus.
